چکیده

Angular light-scattering studies have been carried out on suspensions of isolated rat liver mitochondria. The angular scatter pattern has a large forward component, typical of large particles. Changes in dissymmetry and in the intensity of light scattered at 90 degrees have been correlated with changes in optical density during the course of mitochondrial swelling and contraction. Such changes can be measured at mitochondrial concentrations much below those required for optical density measurements. Changes in mitochondrial geometry caused by factors "leaking" from mitochondria, not detectable by optical density measurements, have been demonstrated by measuring changes in dissymmetry. Angular light-scattering measurements therefore offer the advantages of increased sensitivity and of added indices of changes in mitochondrial conformation.

Contribution of the mitochondrial compartment to the optical properties of the rat liver: a theoretical and practical approach.

The purpose of this work was to analyze the contribution of the mitochondria to the optical properties, i.e., light absorption and scattering, of the blood-free rat liver. Firstly, a theoretical model of the reduced scattering coefficient of the liver was performed by using the Mie theory, the Rayleigh-Debye-Gans approximation, and the electron microscopy descriptions of the liver ultrastructur...

Light scattering from intact cells reports oxidative-stress-induced mitochondrial swelling.

Angularly resolved light scattering measurements were performed on suspensions of EMT6 cells and on mitochondria isolated from rabbit liver. Mie theory analysis of the scattering from intact cells indicated that mitochondrial-sized organelles dominated scattering in the range 5-90 degrees . This interpretation was supported by the analysis of scattering from isolated mitochondria. Intact cells ...

SIZE AND SHAPE TRANSFORMATIONS CORRELATED WITH OXIDATIVE PHOSPHORYLATION IN MITOCHONDRIA II. Structural Changes in Mitochondrial Membrane Fragments

It has been demonstrated that the nature of the physical change in mitochondrial membrane fragments associated with the action of the respiratory enzymes is likely one of shape or symmetry rather than size. The findings suggest that in the state of decreased scattering the macromoleculcs may be present in an extended physical state. Conditions favorable for phosphorylation may givc rise to a fo...
